ABSTRACT:
By creating phytosomes from guava (Psidium guajava) leaf extract and introducing them into mucoadhesive buccal patches, the current study aims to provide a novel drug delivery method. Bioactive phytochemicals with potent antibacterial, anti-inflammatory, and antioxidant properties, including flavonoids, tannins, and terpenoids, are found in guava leaves. Despite these therapeutic benefits, guava leaf extract's low water solubility and limited permeability across biological membranes, which lead to low bioavailability, frequently limit its clinical utility. Phytosomes, lipid-based vesicular delivery systems that enhance the solubility, permeability, and stability of compounds originating from plants, were used to encapsulate the extract in order to get around these problems. Phospholipids were successfully used to create the phytosomes, which were then examined for their zeta potential, particle size, and encapsulation efficiency. In order to guarantee adhesion to the buccal mucosa and extended drug release, these phytosomes were subsequently implanted in mucoadhesive buccal patches composed of suitable polymers such as HPMC and carbopol. The final buccal patches were examined for physicochemical characteristics, including moisture content, surface pH, thickness, weight uniformity, and folding durability. Additionally, the formulations' mucoadhesive strength and in vitro drug release kinetics were evaluated. The outcomes demonstrated enhanced stability of the encapsulated extract, controlled drug release over a number of hours, and improved mucoadhesion. All things considered, this work offers a novel platform that could improve the therapeutic potential of guava leaf extract for both local and systemic therapy applications, making it a promising method for the buccal delivery of phytotherapeutic chemicals.
