ABSTRACT:
The intent of this endeavor was to evolve aceclofenac emulgel which can be used as vehicles for the topical drug delivery of aceclofenac and impact of permeation enhancers on the aceclofenac emulgel. The main objective of a topical medication delivery method is to effectively transport the drug across the barrier and diffuse it into the skin. Emulgel combines emulsion with gel, with emulsions delivering hydrophobic medicines and gel enhancing diffusion, spreadability, and stability of the emulsion. Emulgel is a recently developed innovative drug targeting technology that has been extensively researched for controlling the simultaneous release of gel and emulsion. Emulgel formulations provide the most prominent delivery of hydrophobic drugs to their target sites. This study attempted to determine the optimum concentration of penetration enhancer required to develop and assess an emulgel of aceclofenac by using polymer Carbopol 940P and oil phase sesame oil. Different penetration enhancers were used like propylene glycol and isopropyl myristate and effect of the penetration enhancer on the release has been studied. Six different formulations were formulated by changing the concentration of penetration enhancer, and their quality parameters were evaluated. After that, the emulgel's physical properties, pH, spreadability, viscosity, drug content, drug release, steady-state flow, and kinetics were evaluated. The receptor medium used in the in-vitro release investigation was phosphate buffer pH 7.4. Released study indicate drug release from the formulations with varying concentration of penetration enhancers. Formulation G3 shows maximum drug release 93.91% in 210 min and was selected as optimized formulation due to higher amount of isopropyl myristate used as penetration enhancer. Flux, kp of Aceclofenac in the presence of enhancers was measured. The skin irritation test shows no skin irritation. The formulations were found to be stablein regards to theirphysical characteristics, pH, drug content, and drug release at room temperature and conditions for three months.
