ABSTRACT:
Background: Vulvovaginitis is a prevalent gynaecological condition. Along with effectiveness, patient compliance and ease of use are of primary concern in its management. Although Ayurveda offers variety of remedies (oils, pastes etc.), their contemporary suitability is debatable. Thus, in present study an herbal vaginal gel (MRUDUL) was developed considering Ayurveda principles. Aim: The aim was to formulate and characterize a vaginal gel using Ayurveda ingredients that is both efficacious and convenient, and also prove its activity potential against common vulvovaginitis pathogens. Methodology: A medicated oil was prepared from eight herbal ingredients with known antimicrobial and anti-inflammatory actions viz., Neem, Lajjalu, Haridra, Yashtimadhu, Udumbara, Triphala, and Tila Taila. A gel was developed with this medicated oil as base. The phytochemical analysis, HPTLC fingerprinting, physicochemical characterization, microbial load testing, and accelerated stability assessment were done. In-vitro antimicrobial activity was tested using a time-kill assay against Gardnerella vaginalis and Candida albicans. Results: The final gel formulation had an acceptable pH suitable for vaginal mucosa, high viscosity, good spreadability, and homogeneity. The HPTLC substantiated similar phytochemical presence in oil and gel. No microbial contamination or phase separation of gel was observed for 12 months. The antimicrobial activity study showed >99.9% reduction within 12 hours for both micro-organisms. Conclusion: The MRUDUL gel, rooted in Ayurveda and formulated through contemporary pharmaceutics, demonstrated promising in-vitro antimicrobial activity and excellent stability. It offers patient-compliant alternative with minimal additives and maximum natural ingredients for vulvovaginitis management.
