ABSTRACT:
Mangrove plants grow widely along coastal areas in Sigandu mangrove forest, Indonesia and are often used as medicinal plants by the society, but they remain scientifically underexplored. This is the first study which assess the bioactive compounds, antioxidant, and anti-inflammation activity of mangrove plants from Sigandu, Indonesia. We identified 5 mangrove species based on their morphology, including Rhizophora apiculata, Rhizophora mucronata, Avicennia alba, Avicennia marina, and Bruguiera gymnorrhiza. Mangrove leaves were extracted using ethanol 90%. The extracts were analyzed through phytochemical screening. The DPPH (1,1-diphenyl-2-picrylhydrazil) assay was used to evaluate their antioxidant potential. The anti-inflammatory potential was assessed by examining its impacts on carrageenan-decrease paw edema in rats. Interleukin (IL)-1ß and tumor necrosis factor (TNF)-a concentrations in the inflamed paw were determined using ELISA kits. The findings showed that all extracts contained flavonoids, phenolics, terpenoids, tannin, and only Rhizophora apiculata did not contain saponin compounds. The antioxidant activity of five extracts were very strong, with Bruguiera gymnorrhiza being the strongest with IC50 value of 6.74 µg/mL. In vivo study showed that Avicennia alba provided the most significantly different (p<0.05) paw edema thickness compared to the negative control. Similarly, we found that five species of mangrove leave significantly lowered the concentration of pro-inflammatory cytokines. Moreover, Avicennia alba at dosage 400 mg/kg BW possessed the ability to reduce the concentration of TNF-a inside the inflamed tissue as effective as ibuprofen as drug standard. Thus, the ethanolic extracts of mangrove leaves from Sigandu are potential source of antioxidant and anti-inflammatory agents, which may be caused by secondary metabolites such as flavonoid, phenolics, terpenoids, and tannin.
