ABSTRACT:
Introduction. Coriander (Coriandrum sativum L.) has been used for many centuries as a spice and as a medicinal plant, monographs on the fruits and essential oil of which are included in the leading pharmacopoeias of the world. Coriander sativum herb is promising for use in medicine, it has a large number of important pharmacological effects, including those relevant for the treatment of socially significant diseases. It is necessary to develop regulatory documentation for quality control of a new pharmaceutical plant substance - coriander sativum herb. For this purpose, we studied the identification characteristics, the composition of biologically active substances (BAS) and the antioxidant activity of coriander sativum herb. Materials and methods. The object of the study was dried whole coriander herb, cultivated in temperate and subtropical geographical zones. The morphological and anatomical study was carried out in accordance with the requirements of the State Pharmacopoeia of the Russian Federation XV edition GPM.1.5.1.0002 "Herbs" and GPM.1.5.3.0003 "Technics of Microscopic and Micro-chemical Research of Medicinal Plant Raw Drugs" using optical and luminescent microscopy. Preliminary detection of BAS was provided by generally accepted qualitative reactions. The quantitative content was determined according to the SPRF XV ed. methods: total flavonoids – by the spectrophotometry after the complexation reaction with aluminum chloride, total tannins - permanganometry in terms of tannin, ascorbic acid - by titration with 2,6-dichloroindophenol. Antioxidant activity was determined spectrophotometry after the inhibition reaction of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. Results and discussion. The morphological and anatomical diagnostic features of coriander sativum herb were studied. The structure of the epidermis, mesophyll and vascular system was characterized. It was found that the variety and cultivation area do not affect the manifestation of features. It was determined that tannins in terms of tannin are the dominant substances and make up from 1.71±0.05% to 5.98±0.23%. Also, the content of flavonoids in terms of rutoside was found in coriander herb and amounted from 0.373±0.010% to 1.084±0.015% and ascorbic acid - from 4.60±0.16 to 16.10±0.70 mg%. Determination of the antioxidant activity IC50 DPPH showed a result from 0.00105±0.00003 mg/l to 0.14100±0.00596 mg/l. Conclusion. As a result of the study, data of the chemical composition of coriander herb was obtained and diagnostically significant external features and anatomical structure were determined. This allows to establish the identification of plant raw materials in order to make regulatory documentation for a new herbal drug type.
