ABSTRACT:
This study investigates the cytotoxic and wound healing properties of ethanol, ethyl acetate, and n-hexane extracts derived from the petiole of Colocasia gigantea (Hook.f) on T47D breast cancer cells. The hypothesis assumes that these extracts contain active secondary metabolites contributing to cytotoxicity and cell migration modulation. The cytotoxic activity was measured using MTT assay with five concentrations (31.25 to 500 µg/mL), and IC50 values were calculated. The wound healing assay (scratch method) was performed using non- toxic concentrations (31.25µg/mL). Phytochemical constituents were identified using standard tests based on Farmakope Indonesia VI. The IC50 values were 8.52µg/mL (n-hexane), 21.62µg/mL (ethanol), and 51.11µg/mL (ethyl acetate). Compared to doxorubicin (IC50 ˜ 0.75µg/mL), n-hexane extract showed strong cytotoxicity. Ethyl acetate extract showed highest wound closure (61% at 48h) versus Bionect® control (56%). Ethanol and n-hexane extracts exhibited weak wound healing activity (26% and 25%, respectively). Phytochemical analysis confirmed flavonoids, phenolics, and saponins in ethyl acetate extract, potentially responsible for its pro-migratory effects. C. gigantea petiole extracts demonstrate differential bioactivity: n- hexane exhibits potent cytotoxicity while ethyl acetate extract promotes wound healing, suggesting potential for targeted therapeutic applications.
