ABSTRACT:
The present study highlights the development and validation of a robust and highly sensitive liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the simultaneous estimation of teneligliptin and rosuvastatin in biological matrices. We underwent optimising the method to achieve efficient methodical conditions for analysis, assuring high separation, resolution which ultimately yields good selectivity, accuracy, and reproducibility. The chromatographic separation was performed on a Luna Phenyl-Hexyl column (250mm × 4.6mm, 5µm), employing an acetonitrile-buffer (50:50 v/v) mobile phase at a constant flow rate of 1.0mL/min under isocratic conditions and detected using a SCIEX QTRAP 5500 mass spectrometer operated in positive electrospray ionization (ESI) mode, with multiple reaction monitoring (MRM). The method demonstrated excellent elution with retention times of 2.4min and 4.08min, respectively. The analytes were protein precipitated with acetonitrile for recovery. Method validation was aligned to the ICH Q2 guidelines. Its successful application in pharmacokinetic studies demonstrates its potential for practical utility in clinical research and for routine bioanalytical applications.
