ABSTRACT:
The study aims to formulate and assess floating tablets of pioglitazone employing natural gums like neem gum, in contrast to synthetic cellulose derivative such as HPMC K4M as matrix formers. In order to help preserve buoyancy, sodium bicarbonate was introduced as gas generating agent, which generated CO2 in the gastric acid environment. Addition of bees wax has significantly enhanced the buoyancy of the tablets formulated with both polymers. Gelatin which is used as binder provides strength to the tablet. Through the use of direct compression, the pioglitazone floating tablets were prepared. Precompression parameters, physical attributes, in vitro release, floating duration, and floating lag time were all assessed for the produced tablets. The formulation was optimized for varying neem gum and HPMC concentrations. In vitro release studies revealed that optimized formulation (F13) could release the drug (98%) for 12hrs while maintaining its buoyancy for the same amount of time.
