ABSTRACT:
The current investigation focuses on the synthesis, physicochemical characterization, and biological screening of innovative Azetidin-2-one scaffolds as prospective antidiabetic agents. All synthesized compounds adhered to Lipinski’s Rule of Five and demonstrated favorable drug-likeness and pharmacokinetic profiles through in silico predictions. Among the series, compound 35 displayed potent in-vitro inhibitory effects on enzyme a-amylase, a-glucosidase, and aldose reductase, in addition to high docking scores against enzymes implicated in diabetes, particularly GSK-3ß. In vivo studies in STZ-induced diabetic rats confirmed its significant glucose-lowering effect, improved insulin levels, lipid profile normalization, and enhanced liver glycogen content. Acute oral toxicity testing indicated a high safety margin (LD50 > 2000mg/kg). Compound 35 shows multifunctional potential for type 2 diabetes based on computational, biochemical, and pharmacological data.
