ABSTRACT:
The present study aims to produce and validate easy, accurate, precise, inexpensive, rapid and sensitive UV-spectrophotometric technique to determine Berberine and Gallic acid simultaneously in polyherbal churna. Method was developed and validated in terms of LOD, LOQ, robustness, accuracy and roughness. The developed method was used for determination of Berberine and Gallic acid in polyherbal churna. The maximum absorbance of the drugs was found at 229nm for Berberine and 217nm for Gallic acid, respectively. The linearity response concentration range (10-50µg/mL) of Berberine and Gallic acid was observed with ( Y = 1.9577x +0.0184, r2 =0.998 and Y = 1.8506x +0.0028) respectively. The accuracy was found between 98.25% and 101.9%. The precision for intra and interday measurements were found to be within the limits. LOD and LOQ were calculated to determine the method's sensitivity and the values were found to be 0.014µg/mL and 0.042µg/mL for berberine, gallic acid's values were found to be 0.012µg/mL and 0.038µg/mL, respectively. The suggested approach has been verified and can be applied to the regular examination of Berberine and Gallic acid in pharmaceutical dosage forms.
