ABSTRACT:
Background: Burn injuries pose a significant health concern and can lead to severe complications. It is essential to promote wound healing and minimize scarring, which are the most common forms of injury. Due to their broad availability and extensive use in traditional healing practices, herbal remedies are often favoured for managing various health conditions. Pachai Pakku Thailam (PPT) is one such traditional formulation that has long been employed in the treatment of skin ailments and wounds. Objectives: The first research of this study focuses on evaluating the wound healing potential of Pachai Pakku Thailam in full-thickness burn injuries using a Wistar rat Model. Methodology: Wistar rats were assigned to three groups: Group I (control) and Group II (Treated with Silver Nitrate ointment) and Group III (Treated with PPT). Burn injuries were induced, and topical treatments were applied daily for a duration of 42 days. Biochemical markers such as total protein, hydroxyproline, hexosamine, and hexuronic acid levels were analyzed. Wound contraction was evaluated at different day intervals, and a histological analysis was performed on the 7th day and the 42nd day. Results: From the 7th to the 42nd day, the PPT group exhibited significantly enhanced wound contraction in the early stages (Day 7) compared to the control group. The PPT group showed increased collagen synthesis, improved tensile strength (908.06 µg/g), and remodeling. The histological study reveals a dense matrix of metalloproteinase, inflammatory infiltrates, fibroblasts, with thin-walled congested vessels formed on the 7th day, and normal epidermis with dermis-congested blood vessels and fibrous collagenous congested vessels on the 42nd day. This expression also confirmed the healing efficacy of Pachai Pakku Thailam. Conclusion: The findings prove that the Pachai Pakku Thailam Siddha formulation significantly improves the wound healing phase, collagen synthesis, tensile strength, and tissue regeneration.
