ABSTRACT:
Verapamil HCl, a calcium channel blocker with a short half-life, requires frequent dosing for effective hypertension management. To enhance therapeutic efficacy and patient compliance, a capsule combining immediate-release (IR) and extended-release (ER) pellets was developed and optimized to achieve both rapid onset and prolonged drug release. Drug layering was performed on spheres followed by protective coatings using Eudragit and ethylcellulose, with a 3² full factorial design employed to optimize polymer concentrations. The formulation was evaluated through DSC, FTIR, SEM, flow properties, friability, assay, in vitro dissolution, and stability studies. The optimized batch (F4) demonstrated biphasic release with an initial 20% release within 2 hours and a cumulative release of 96% over 24 hours, following first-order kinetics with concentration-dependent release. FTIR and DSC confirmed drug–excipient compatibility, SEM revealed intact pellet coatings, and the formulation exhibited suitable mechanical and flow properties for capsule filling. Stability under accelerated conditions was maintained for five months. These findings indicate that the developed IR-ER Verapamil HCl capsule offers improved patient compliance and therapeutic efficacy in hypertension management.
