ABSTRACT:
Tuberculosis is caused by Mycobacterium tuberculosis and affects individuals irrespective of age, sex, or socioeconomic status. Monitoring IgM and IgG antibody levels during therapy is essential to identify latent and active tuberculosis. Coleus scutellarioides and Phyllanthus niruri have been traditionally used as immunomodulators and antituberculosis remedies; however, scientific validation, particularly regarding antibody profiles during treatment, is still limited. This study aimed to evaluate the changes in IgM and IgG levels during tuberculosis infection following intervention with miana and meniran extracts. The hypothesis proposed that Miana Leaf Extract (MLE) and Meniran Herb Extract (MHE) can reduce IgM and IgG levels as immune responses during tuberculosis infection through immunomodulatory mechanisms. Methods: MLE and MHE were prepared using maceration. Mice were infected with M.tb and incubated for seven days. The animals were allocated into four groups: K1 (MLE), K2 (MHE), K3 (Antituberculosis Drugs/ATD), and K4 (placebo). Blood samples were collected on days 0, 14, 21, and 28 to determine IgM and IgG levels by ELISA. Results: IgM and IgG expression reflected the infection status. Pre–post treatment analysis showed a decrease in ?IgM and an increase in ?IgG after 28 days in MLE (135.56/126.43), MHE (166.60/105.04), ATD (110.71/46.38), and placebo (338.90/273.99) groups. Mann–Whitney analysis (a=0.05) indicated ?IgM differences of MLE–MHE (p=0.437), MLE–ATD (p=0.324), MHE–ATD (p=0.829), and significant differences for MLE/MHE/ATD vs. placebo (p=0.001–0.006). ?IgG values showed MLE–MHE (p=0.322), MLE–ATD (p=0.002), MHE–ATD (p=0.017), and significant differences for MLE/MHE/ATD vs. placebo (p=0.000). Conclusion: Miana and meniran demonstrated potential immunomodulatory effects during the early phase of tuberculosis infection, resulting in greater reductions in IgM and IgG levels compared with placebo.
