ABSTRACT:
Aim: Gentamicin is an aminoglicoside antibiotic that has nephrotoxicity and hepatotoxicity effect. This study was conducted to detemine the activity on roselle extract to protect Liver an Renal from Gentamicin toxicity. In addition, to understand the protective effect of roselle extract due to gentamicin toxicity on liver and renal in rat as a model animal by assessing the AST, ALT and histopatology of liver and renal. Material and Methods: There were 25 rats utilized in this inquire about which were separated into five bunches. Control bunch (K-) were given 0.5% CMC-Na and aquadest, the positive control gather (K+) were actuated by Gentamicin 80 mg/kgBW, and the treatment gather (P1, P2, P3) actuated by Gentamicin 80mg/kgBW and Roselle blossom extricate by changing the dosage (200mg/kgBW, 400mg/kgBW, 600mg/kgBW). On day 16, all of the white rats were anesthetized and necropsied. Their blood was taken by intracardiac for ALT and AST value test using spectrophotometer, the results were analyzed using the One-Way Analysis of Variance (ANOVA) was followed by Duncan’s Multiple Range test as a post hoc test if the analysis showed a significant result (p<0.05). ; the kidney and liver organs were taken for infinitesimal examination with Hematoxylin Eosin recoloring, the comes about were analyzed utilizing the Kruskal-Wallis test that demonstrates a noteworthy contrast (p<0.05), and the contrasts between medications were appeared based on the result of the Mann-Whitney test. Result: In this study, based on the results from all bunches showing that Roselle extract was able to decrease the AST and ALT value of white rats after being induced by Gentamicin; based on Roselle extract’s effectiveness as an antioxidant, it protects liver by inhibiting cytochrome P450 chemicals, smothering proapoptotic protein expression and expanding the rate of hepatocyte vitability; it moreover secures kidney by anticipating oxidative push, expanding antioxidant proteins, and repressing pro-inflammatory cytokines. Conclusions: Giving roselle extricate at dosages of 200, 400, and 600mg/kgBW had an impact in progressing the liver harm which was stamped by a diminish within the ALT and AST esteem and degree of harm based on the scoring result within the frame of degeneration, rot, entry irritation, and fibrosis; it is additionally moving forward the kidney harm which was checked by a decrease within the degree of harm based on the scoring result within the frame of degeneration, corruption, and irritation on the proximal tubules in kidneys of rats.
