ABSTRACT:
Yogaraja guggulu is an important classical polyherbal formulation used in the Ayurvedic medicine system for treating various disease conditions like Vatavyadhi (neurological and musculoskeletal disorders) and sandhigata vata (osteoarthritis). The present study was aimed to assess the safety of Yogaraja guggulu through acute and 90-day repeated oral dose toxicity testing. In the acute toxicity study, the test drug was orally administered at a dosage of 3900mg/kg to Swiss albino mice. For the 90-day repeated dose oral toxicity study, three different doses of the test drug (270mg/kg, 1350mg/kg, and 2700mg/kg) were administered to Wistar rats. During the study, observations were made on cage-side behaviour, body weight, and feed intake. No abnormal behavioural activity or pre-terminal deaths were observed in mice exposed to a single dose of 3900mg/kg. There were no pre-terminal deaths in rats receiving the test compound at three different doses for 90 days. There were no significant changes in haematological and biochemical parameters and histopathological analysis of major organs revealed no major lesions or treatment-related changes. During the present study, Yogaraja guggulu was found to be safe up to 3900 mg/kg in mice upon single exposure and up to 2700mg/kg upon 90 days of consecutive oral administration in rats.
