ABSTRACT:
The observation and design of direct floating gastroretentive beads (DFGB) through emulsification of herbal extracts in polymer hydrogels is an effective alternative to overcome the weaknesses of floating gas generating system. The aim of this research is to investigate and design gastroretentive a direct floating combination of ethyl cellulose with a hydrophilic polymer containing turmeric extract and Centella asiatica extract to treat stomach ulcers. DFGB was prepared by modifying the solid dispersion technique using the solvent evaporation method, the liquid dispersion was prepared by emulsification herbal extracts in various combinations of hydrophilic polymer hydrogels, using tween 80 and span 80 as emulsifying agents and 5% ethyl cellulose as a buoyancy enhancer. The dispersion liquid mixture was observed for floating properties and the particle size distribution was measured using a Particle Size Analyzer (PSA) before being molded into solid beads. The gastroretentive characteristics of the bead preparation were investigated in vitro by observing the onset of floating, duration of floating, swelling properties, and integrity of the beads in the artificial stomach medium. Emulsification of turmeric and Centella asiatica extracts in a polymer hydrogel produced a liquid dispersion with floating properties, a homogeneous particle size distribution at the nanoscale, and was ready to be printed. The combination of alginate-pectin polymer with 5% ethylcellulose shows that several formulas including F2, F5, F6, and F9 have immediate floating properties from zero minutes (without lag time ) with a floating duration of more than 12 h, swelling, and slow erosion over time >12 h. The formulation of turmeric extract and Centella asiatica extract emulsified in a polymer hydrogel combination of sodium alginate pectin and 5% ethylcellulose solution in ethanol has the potential to be developed as a direct floating gastroretentive system for the treatment of gastric ulcers.
