ABSTRACT:
Objective: Homeostasis is a major factor in the field of biology and medical sciences. The study of mechanism of clock genes and the reason behind 24 hr sleep wake cycle is called chronobiology. The gap between the action of medicine and rhythmic variation can be effectively bridged by chronopharmaceutical drug delivery system. Formulation where constant drug release is not required chronopharmaceutical drug delivery system is preferred. The risk factor for patients suffering with cardiovascular diseases are more in early morning as the blood pressure rises in a significant manner just before awakening hours. The purpose of present study was to prepare chronopharmaceutical drug delivery system for propranolol hydrochloride by film coating method. Method: Film coating method involves polymers like eudragit L100 and eudragit S 100 as coating agent in the ratio1:1. The coated tablets were evaluated for uniformity of thickness, in vitro release and release kinetics study. Results: When investigated the tablets coated with different thickness of eudragit L 100 and S 100 received the weight gain up to 12%. In vitro release study found that the coating thickness of 0.25mm resulted in complete resistance to the PH of stomach and intestine maintaining a lag time of 6 hr. The release kinetics was interpreted to follow higuchi model. Conclusion: The above study was successfully designed to prepare chronopharmaceutical drug delivery system of propranolol hydrochloride by film coating method.
