The ever-growing need and greed of human beings have resulted in increased stress, mental disorders and psychological misbehaviors are the outcomes of so-called modernized life. Varying mental disorders are taking millions of people under its wings every year, so is the ever-expanding need of curing it. Many drugs are the choices for treatment and effective management of psychosis; Quetiapine fumarate is one of them. Because of the bioavailability and gastrointestinal concern which makes its journey into the body difficult, a lot has to be done to make it a perfect choice. To achieve spatial (site) control over the drug release, sustained release formulations are useful to release drug at specified site so as to ease its absorption. Buccoadhesive drug delivery systems include any drug delivery systems that achieve slow release of drug in buccal cavity, getting absorbed from the buccal mucosa, though way protecting itself from the harsh gastric environment and keeping plasma drug concentration consistently at desired level for an extended time. In the present study, work has been done to formulate and evaluate sustained release buccoadhesive matrix tablet containing Quetiapine fumarate, a blend of polymers including HPMC K4M and K15M, Carbopol 934P, xanthan gum polyvinyl pyrrolidone K30. Wet granulation method is used for tablet formulation. Preformulation and post-formulation evaluation studies were performed on the drug-polymers blend as well-prepared tablets in relevance to various parameters. Study and test results as FTIR analysis proved compatibility of polymers with the drug; the blend of polymers help control the drug release over an extended time period. Bioadhesive studies revealed promising adherence to buccal mucosa helping for controlled drug release and thereby enhanced bioavailability. The release analysis revealed erosion mediated drug release.
