ABSTRACT:
Amphotericin B (AmB) is a polyene macrolide antibiotic which shows antifungal activity against the widest spectrum of fungal infections and in certain protozoan infections with no major problems of resistance and remains effective till date. Literature has given insight on the preparation of different novel formulations of AmB as lipid formulations and polymeric nanoparticles using UV-VIS spectrophotometry and HPLC method, but details of peak characteristics and validation data are not described. This research was focused to develop a suitable validation method of AmB and acquire consistent, reliable and accurate data on linearity, detection limit, quantification limit, accuracy and precision using a UV-VIS spectrophotometer. The UV-VIS spectrum of AmB was recorded with the study of linearity, accuracy, precision, detection and quantification limit was carried for the validation of the process. The method showed good linearity with regression of y =0.1355x (R2 = 0.9992) at 408 nm. The repeatability (inter-day) and intermediate precision (intra-day) precision studies revealed that the method is precise and reliable where all the RSD values were ?2%. Method accuracy showed % accuracy value for all the three concentration levels ranged from 103.5% to 106.5% with standard deviation ranging from ± 0.763 to ± 1.12 that a small change in the concentration of the drug could be accurately determined with high accuracy. Limit of detection and limit of quantification were 0.391 (µg/ml) and 1.187 (µg/ml) respectively. The proposed method is simple and could be regarded as a reasonably feasible technique in the routine estimation of AmB.
