ABSTRACT:
Enantiomeric separation of metoprolol tartrate was developed using a reversed phase HPLC (RP-HPLC) with methyl-??-cyclodextrin (M-??-CD) as a chiral mobile phase additive. The effects of the pH value of mobile phase, concentration of chiral additive, composition of mobile phase on the enantioseparation of metoprolol tartrate were investigated on Macherey Nagel C18 column (250 mm×4.6 mm, 5.0 µm). The detection was performed using UV at 274 nm. A satisfied resolution was achieved using a mobile phase consists of a mixture of aqueous solution( 1.5g/l M-??-CD), and methanol with a volumetric ratio of (86/14)v/v%, and flow rate of 0.5 mL/min. The linearity, precision and accuracy were studied to prove the validity of the developed method and could simply applied to determine the enantiomers of metoprolol tartrate in pharmaceutical tablets.
