ABSTRACT:
The aim of this work was to improve the invitro dissolution of Simvastatin through development of self nanoemulsifying capsules. Simvastatin is a poorly-water soluble drug and cholesterol lowering agent. It has poor oral bioavailability (5%) due to its low solubility. So self nanoemulsifying drug delivery of Simvastatin increases its solubility and enhance its bioavailability. Nanoemulsions of ternary phase diagrams constructed usingvarious modified oils, Smix (mixture of surfactant and co-surfactant) were used to prepare different self-nanoemulsifying drug delivery systems (SNEDDS). Whose composition was optimized using drug-solubility, system stability and droplet size distribution studies. Optimized SNEDDS were subjected to Characterization studies. Results revealed that systems with 10% relatively polar oils ,Smix (5:1) that means 60% Cremophore® RH 40 (surfactant), and 30% Ethanol(co-surfactant),acquired good self-nanoemulsification properties. The optimized SNEDDS Characterized by droplet size(100nm). In vitro release of Simvastatin was significantly higher when compared with marketed tablets of Simvastatin (Zocor®).Scanning electron microscopy (SEM) study confirms that the droplet size of the SNEDDS With in nanoscale.It can be concluded that the optimized formula were able to introduce Simvastatin successfully in SNEDDS to enhance the oral bioavailability of Simvastatin.
