ABSTRACT:
A accurate, precise, specific modified High Performance Liquid Chromatography method was developed for the simultaneous quantification of Bupropion and Naltrexone in bulk and dosage forms. A C18 column (250 x 4.6mm; 5 µm Phenomenex) with green mobile phase containing 0.05% v/v N,N-Diisopropylethylamine (DIPEA) (pH6.5): water (25:75% v/v) 20°C was used and isocratic pump is used for elution and eluents were monitored at 251 and 281 nm. The retention times of bupropion and naltrexone were 2.692 min and at 4.56 min respectively and showed a good linearity in the concentration range from 1?g/mL to 200 ?g/mL of Bupropion and Naltroxone were prepared and detected by HPLC. Concentrations of Bupropion and Naltroxone 251 nm were found to be linear in the range of 5?g/mL-40?g/mL and 10?g/mL-90?g/mL respectively with a correlation coefficient of 0.98and 0.998 respectively. The average percent recoveries were found to be 99% for bupropion and naltrexone. The developed method fallows all the validation parameters like accuracy, precision, linearity, limit of detection, limit of quantitation and solution stability. Conclusion: The proposed method was validated as per International Conference on Harmonization guidelines and successfully applied to the simultaneous estimation of Bupropion and Naltrexone in bulk and dosage forms.
